Depends on the vacuum. Few millimeters of mercury can be measured using the mercury barometer, or a bourdon tube gauge etc. Medium vacuum is measured using more sophisticated gauges like thermocouple gauges etc., Very high vacuum is measured using still more sophisticated instruments.

Well... a vacuum is a space in which there are few, or no particles. So, there are only a couple of things you can measure about a vacuum, like: volume (of the total area of vacuum), density (of any particles in the vacuum), pressure, and temperature.
